Agar detergente semisólido: una alternativa para la conservación ex situ de Cladosporium spp. (Ascomycota: Cladosporiaceae)

Abstract

El género Cladosporium es ubicuo y tiene una amplia representación ambiental. Para la conservación de cepas de este género fúngico se han utilizado varios métodos de elección, entre los que se encuentran la criopreservación y la liofilización. Sin embargo, existen otros métodos alternativos y complementarios que disminuyen los costos de conservación. El objetivo de este trabajo fue evaluar la factibilidad de la conservación de 92 cepas de Cladosporium en el medio Agar Detergente Semisólido (agar bacteriológico al 0,2 %, Tween 80 al 0,05 % y 100 ml de agua destilada estéril). Se determinó viabilidad, pureza e integridad morfológica hasta tres años posteriores a la conservación inicial. Para la conservación se utilizaron tubos Eppendorf™ de 1,5 ml con 1 ml de medio, a los que se inocularon esporas obtenidas a partir de un cultivo de 7 días en Papa Dextrosa Agar y se mantuvieron a 4 °C. Durante el período evaluado, las cepas mantuvieron la viabilidad con valores superiores al 90 %, por lo que este puede ser un método alternativo para la conservación de cepas de Cladosporium a corto o mediano plazo.
