Ruptura mecánica de la levadura Pichia pastoris para la extracción de la proteína recombinante VP60

Abstract

El sistema de expresión de la levadura metilotrófica Pichia pastoris ha sido ampliamente empleado en la obtención de proteínas recombinantes con fines terapéuticos, entre las que se encuentra la proteína VP60 como principio activo del candidato vacunal contra la fiebre hemorrágica del conejo. Esta proteína es obtenida de forma soluble e intracelular por lo que precisa ser extraída para su posterior purificación. En la literatura se emplea básicamente la ruptura mecánica empleando molinos de bola para este tipo de sistema, sin embargo en este caso específico no se conocen las condiciones de operación de esta etapa del proceso, por lo que el objetivo de este trabajo es establecer las condiciones de operación de la etapa de extracción de la proteína VP60. Para ello se estudiaron los tiempos de retención combinados con los diferentes número de pases, además se establecieron otros parámetros de trabajo como la carga de perlas de vidrio en la cámara de molienda, el diámetro de las perlas, el flujo de trabajo, la concentración celular de la crema, la velocidad del rotor de agitación y el pH de la crema empleando equipos de laboratorios e industriales. Los experimentos realizados permitieron concluir que la ruptura mecánica con molinos de bolas logra extraer de manera eficiente la proteína VP60 con 5 pases de ruptura de un min cada uno y los resultados obtenidos resultan similares a los reportados para la obtención de otras proteínas en este sistema de expresión.
