Single Cell Transcriptomes From Mouse Cerebellum

Abstract

species : Mus musculus sex : male strain : C57BL/6J age : post-natal day 40 P40, mouse cerebellum single cells were obtained by Papain dissociation (Worthington), and neuronal enrichment was achieved by Percoll gradient selection. Samples were loaded onto a C1/Fluidigm and processed using an adapted Smart-seq2 protocol (adapted from Picelli et al Nature Methods 2014). Each sample/well also contains 92 ERCC spike-ins in a concentration of 1/40000 in the lysis buffer. The amplified cDNA libraries were prepared for sequencing using the Nextera XT DNA sample preparation protocol adapted for single cell followed by sequencing on an Illumina HiSeq machine.
