José Gregorio Hernández Cisneros nace en la población de Isnotú, estado Trujillo, Venezuela, el miércoles 26 de octubre de 1864, hijo de Benigno Hernández Manzaneda y de Josefa Antonia Cisneros Monsilla (el primogénito de seis hijos). Su madre muere el 18 de agosto de 1872, por lo que, con apenas 8 años, José Gregorio queda bajo la protección de su tía materna María Luisa. En Isnotú inicia sus estudios para luego trasladarse a Caracas (a los 14 años), era un trayecto muy largo y complejo (en esta época no existía la carretera trasandina), primero se trasportaban en mulas hasta el puerto de “La Ceiba”, luego por vía lacustre hasta Maracaibo, de allí en barco hasta Curazao (presentaban pasaporte) para posteriormente trasladarse a La Guaira y seguidamente atravesar el cerro “Ávila” por el “camino de los españoles” para finalmente llegar a Caracas
