Variabilidad genética en el microsatélite "A" del extremo 3´UTR del gen SLC11A1 en un rebaño de cabras Criollas (Capra hircus)

Abstract

El gen SLC11A1 codifica un transportador catiónico localizado en la membrana del fagolisosoma de los macrófagos, que tiene participación activa en la supresión de infecciones bacterianas y presenta dos microsatélites polimórficos (dinucleótido GT) en el extremo 3’UTR y algunos de sus alelos han sido asociados con resistencia natural a patógenos intracelulares en animales domésticos. A fin de caracterizar la variabilidad genética del microsatélite “A” del extremo 3´UTR del gen SLC11A1 presentes en un rebaño de cabras Criollas localizado en el estado Lara, se amplificó mediante una PCR un fragmento de 233 pb a partir del ADN aislado de 62 muestras sanguíneas colectadas en el centro de producción de ovinos y caprinos del INIA-Lara. Los amplicones obtenidos fueron sometidos a un análisis de polimorfismos conformacionales de cadena simple (SSCP) y las muestras fueron agrupadas según la identidad de su patrón SSCP; de esta manera se formaron seis grupos de análisis. Se tomaron muestras representativas de cada grupo para determinar el número de repeticiones GT en el microsatélite “A” mediante secuenciación de ADN. Las frecuencias genotípicas fueron estimadas a partir de la secuencia nucleotídica correspondiente a cada grupo de análisis (GT12/GT17=0,016; GT12/GT13=0,016; GT13/GT13=0,016; GT13/GT15=0,290; GT13/GT16=0,483; GT16/GT18=0,177) y las frecuencias alélicas obtenidas fueron GT12=0,0161; GT13=0,4113; GT15=0,1452; GT16=0,3306; GT17=0,0081 y GT18=0,0887. El coeficiente de información polimórfica fue 0,6390, la heterocigosidad observada fue 98,39% y la heterocigosidad esperada era 69,79%. El rebaño muestreado no se encuentra en equilibrio genético (P<0,05).
