Abstract

Guión de trabajo para el laboratorio virtual «Cibertorio» en Biomodel.UAH.es La identidad de una infección vírica se puede verificar detectando la presencia del material genético del virus en muestras tomadas al paciente. En concreto, este ensayo se centra en la detección de secuencias propias del RNA genómico de SARS-CoV-2, el coronavirus causante de la pandemia COVID-19 que apareció a finales de 2019 y se extendió por todo el mundo en 2020. La identidad del virus se confirma en comparación con secuencias genómicas del influenzavirus A H1N1, causante de la gripe común que tiene síntomas iniciales similares. Como control para verificar la amplificación por PCR se utiliza un gen humano constitutivo, el de la gliceraldehído-3-fosfato deshidrogenasa Como material de partida disponemos de muestras de cDNA (virtuales) preparado a partir del RNA contenido en muestras tomadas de la mucosa de seis pacientes con síntomas de gripe. Objetivo: identificar, en su caso, la infección empleando varios cebadores para PCR dirigidos a regiones del genoma humano, del virus A de la gripe y del coronavirus.
