Rapamycin-Induced Autophagy for Ganglioside Aggregate Clearance

Abstract

Rapamycin inhibits mTORC1 signaling, leading to ULK1 activation and initiation of macroautophagy. Enhanced autophagic flux could facilitate clearance of GM2 ganglioside storage vesicles and aggregated proteins in Tay-Sachs neurons through lysosomal fusion and bulk degradation mechanisms, bypassing the specific HEXA enzyme deficiency.
