Open Targets identifies CDKN2A with 0.29 association score in EMPD. CDKN2A loss leads to unchecked CDK4/6 activity driving cell cycle progression. CDK4/6 inhibitors like palbociclib could restore cell cycle control. ClinVar shows multiple CDKN2A variants associated with cancer predisposition, suggesting germline testing could identify high-risk patients.
