Antisense oligonucleotides targeting MAPT mRNA would reduce total tau protein expression in PSP-affected brain regions. By decreasing the substrate available for pathological phosphorylation and aggregation, this approach addresses the root cause of PSP tauopathy rather than downstream effects.
