FlexConsensus: Replicación metodológica de un algoritmo de fusión de paisajes conformacionales para cryo-EM

Abstract

Introducción: La criomicroscopía electrónica (cryo-EM) permite visualizar proteínas en múltiples conformaciones, pero distintos algoritmos de IA producen paisajes conformacionales inconsistentes. FlexConsensus (Herreros et al., Nature Methods 2025) resuelve este problema mediante un multi-autoencoder. Objetivo: Replicar metodológicamente FlexConsensus implementando una aplicación web interactiva. Métodos: Se desarrolló una aplicación full-stack (Flask, NumPy, Plotly) que simula espacios latentes de CryoDRGN y HetSIREN, implementa el multi-autoencoder, y calcula el test de Mantel y el consensus error. Resultados: La réplica alcanzó r = 0.793 en el test de Mantel (superando el umbral r > 0.70) e identificó 20.5% de partículas confiables (vs ~20% en el paper original), con p = 0.001. Se detectaron 7 conformaciones para el anticuerpo 1IGY. Conclusiones: La implementación replica exitosamente la metodología central de FlexConsensus, validando que el enfoque de consenso es reproducible. Constituye una herramienta didáctica accesible sin necesidad de GPU.
