PurposeProvides supporting data for the study "Machine Learning and Experimental Validation Reveal Autophagy-Linked Biomarkers in Microvascular Injury after Cardiac Ischemia/Reperfusion", facilitating reproducibility of results. Content Structure1. Bioinformatic Analysis •Directory: /01_Bioinformatics_Analysis/ •Contents: This directory contains the complete results of the bioinformatic pipeline for identifying and validating autophagy-linked biomarkers. It includes: •Differential Expression Analysis: Results from identifying Differentially Expressed Genes (DEGs/). •Functional Enrichment: Results from Gene Ontology (GO/), KEGG pathway (KEGG/), and Gene Set Enrichment Analysis (GSEA/). •Machine Learning for Feature Selection: Code and results from multiple algorithms used to refine biomarker signatures, including Support Vector Machine (SVM-REF/), Random Forest (randomForest/), and LASSO regression (lasso/, lasso-venn/). •Diagnostic Model Building & Validation: Files related to constructing a clinical predictive model, including a Nomogram (nomogram diagram/) and Receiver Operating Characteristic curve analysis (ROC/). •Additional Analyses: Results from immune infiltration analysis (immune/), disease association (disease/), and clustering analysis (cluster/). •Validation Sets: Data and results from the test and validation cohorts (test validation/). •Various Intersection Analyses: Venn diagram results (venn/) comparing gene lists from different methods. 2. Experimental Validation •Directory: /02_Experimental_Validation/ •Contents: Myocardial tissue section staining images (jpg/png), qPCR quantitative data (xlsx), Western blot raw images (TIFF), tunel (TIF), TEM (TIFF). Methodology Data Collection•Bioinformatic Data: RNA-seq data from mouse models sourced from the GEO database (GSE108940, GSE115568, GSE4105, GSE58486, GSE61592, GSE83472) •Experimental Data: C57bl/6J I/R model, sampling time points: ischemia 45minutes and reperfusion 2h (n=4/group); Cardiac Microvascular Endothelial Cells (CMECs) H/R model, sampling time points: hypoxia induction 24h and reoxygenation 24h (n=3/group).
