MLE and LRT Analysis for Single-Molecule koff Estimation

Abstract

Contents `mle_analysis_koff.m` Estimates koff from dwell time data using an exponential decay MLE model. `mle_censoredvsuncensored.m` Compares MLE fits of uncensored vs. right-censored dwell time data. `LTR_for_mle_.m` Performs Likelihood Ratio Test comparing two separate vs. shared koff values. Features Fits single-exponential decay models using MLE.Supports right-censored dwell time data (for photobleaching-limited recordings).Computes standard errors and 95% confidence intervals for koff.Performs Likelihood Ratio Test to assess whether datasets differ significantly. Requirements MATLAB R2014b or newerStatistics and Machine Learning Toolbox

This repository contains MATLAB scripts for analyzing Single Moleclule Pulldown data (SiMPull)- with these scripts, single-molecule dwell time data is used to estimate dissociation rate constants (**koff**) using **Maximum Likelihood Estimation (MLE)**. It also includes tools to compare koff values between datasets using a **Likelihood Ratio Test (LRT)**.
