Identificación de genes homólogos a inhibidores de proteasas presentes en el genoma del oomiceto patógeno de la familia de las solanáceas

Abstract

La secuenciación de genomas de diversos patógenos del género Phytophtora ha conducido a la identificación de genes que codifican proteínas efectoras que permiten la evasión y manipulación de la defensa de la planta. El objetivo de la investigación fue la búsqueda e identificación de secuencias homólogas a proteínas inhibidoras de proteasas y glucanasas, dentro del genoma de P. capsici (CPV-282) aislado de chile en México. La Metodología consistió en analizar inhibidores de proteínas extracelulares (EPI), inhibidores de proteasas con dominios tipo cistatina (EPIC) y proteínas inhibidoras de glucanasas (GIP) reportadas en Phytophtora infestans, Phytophtora sojae, Phytophtora ramorum y Phytophtora brassicae. Resultados obtenidos: El análisis in silico reveló la presencia de genes homólogos a EPI10, EPIC3, PiGIP y PsGIP, a partir de los cuales se diseñaron oligonucleotidos para confirmar la predicción del análisis bioinformático. Las secuencias obtenidas de los productos amplificados por Reacción en Cadena de la Polimerasa permitieron identificar secuencias homólogas a inhibidores de proteasas tipo cistatina (PcEPIC3) e inhibidores de glucanasas (PcGIP), dentro del genoma de P. capsici. Los resultados sugieren que el género Phytophthora comparte herramientas genéticas que le permiten manipular los sistemas de defensa de la planta y así ser eficientes en la colonización de su hospedero.
