Abstract

Bu çalışmada, tarım arazilerinden izole edilen Bacillus spp. suşlarının ürettiği insektisidal proteinler, alan emisyonlu taramalı elektron mikroskobu (FESEM) kullanılarak karakterize edilmiştir. Bacillus thuringiensis (Bt) bakterisinin ürettiği Cry proteinleri, böcekler üzerinde toksik etkiler gösteren kristal yapılardır ve biyolojik mücadele alanında geniş bir kullanım alanına sahiptir. Araştırmada, Kayseri ilinin 5 farklı ilçesinin tarım alanlarından toplanan toprak örneklerinden Bacillus suşları izole edilmiştir. Çalışma kapsamında toplam 203 bakteri izolatı elde edilmiş ve bunların 75 'nin Bacillus cinsine ait olduğu belirlenmiştir. Bu suşlar arasında, 5 Bacillus thuringiensis suşunun insektisidal kristal protein (ICP) ürettiği gözlemlenmiştir. FESEM analizleri sonucunda, bipiramidal, küresel ve düzensiz şekillerde ICP yapıları elde edilmiştir. Bu proteinlerin böcek türleri üzerinde etkili olabileceği ve tarım zararlılarıyla mücadelede biyopestisit olarak kullanılabileceği belirlenmiştir.
