D4.6 Cell analysis and chemical tests results in ALTERNATIVE in vitro system

Abstract

Deliverable 4.6 presents the updates and testing of the sensorized fluidic platform designed within the ALTERNATIVE project and the work on the cell tests with the chemicals selected during the project. The document describes the system's setup for cleaning, sterilization, and assembly of the platform. Experiments were performed to evaluate the impact of different flow rates on cell viability. Furthermore, the potential cytotoxic effect of materials for electrodes and the applied pulse were tested in static conditions. Finally, preliminary data was generated with the nearly complete configuration of the microfluidic platform in the presence of cells cultured in a 3D underflow environment. Regarding chemicals, the document shows the dose-response curves generated in 2D, which help calculate the IC85 dose applied in the chemical tracking experiments for metabolomic analysis performed in 2D and 3D environments.
