In vitro techniques in Solanum lycopersicum, Solanumtuberosum, and Solanum × muricatum anthers

Abstract

Se desarrollaron técnicas in vitro para cultivos Solanum transcendentales, incluyendo propagación a partir de semilla de tomate; micropropagación de papa por medio de brotes de tubérculo; y cultivo de anteras de pepino dulce. En todas las etapas de tomate y papa se usó un diseño completamente aleatorizado (DCA) con o sin arreglo factorial; y se evaluaron diferentes medios de cultivo para anteras de pepino dulce. Se utilizaron tomates Floradade (V1) y Montego (V2), papas Chaucha (CH), Leona Blanca (LB) y Leona Negra (LN), y pepino dulce Nueva Generación (NG), así como dos tratamientos de desinfección (TD), diferentes concentraciones de gentamicina (50, 68.8 y 75 mg · L⁻¹), fungicidas (FF), bacteriostático y bactericida (BB). La germinación de tomate con contaminación < 10% se obtuvo con V1-TD1-68.8 ppm = 86.67%, y V2-TD1-68.8 ppm = 80.00%; mientras que, la longitud del explanto (media ± E.E.) que va desde V2-TD1-68.8 ppm = 12.63 cm ± 0.49, a V2-TD2-68.8 ppm = 3.40 cm ± 1.23 fue significativamente diferente; y la longitud ex vitro de plántula fue diferente entre V1 = 12.64 cm ± 0.68, y V2 = 17.17 cm ± 0.69. En la micropropagación de papa (contaminación 0–6.67% y supervivencia 93.33–100%), el número de hojas fue diferente entre CH-FFBB-75 ppm = 7.60 ± 0.43 (A), LB-FFBB-75 ppm = 4.00 ± 0.58 (B), y LN-FFBB-75 ppm = 1.50 ± 0.27 (C), y la longitud del explanto fue similar, con 2.14, 1.79 y 1.96 cm, respectivamente. Se obtuvieron dos callos a partir de anteras de pepino dulce en medios con kinetina (0.01–0.1 ppm), auxina y citoquinina.
