Abstract

Die Untersuchung des Neisseria meningitidis Adhäsins NadA in Meningokokken ist durch die Anwesenheit diverser Adhäsine erschwert. Daher wurde Y. enterocolitica als Modellorganismus für die funktionelle Charakterisierung des NadA verwendet, und lieferte neue Einblicke in die Interaktion von NadA mit Epithel- und Immunzellen, mit Komplementfaktoren, sowie in die Rolle von NadA bei der Mausvirulenz. Zusätzlich konnte eine spezifische Interaktion von NadA mit humanen β1 und β3 Integrinen nachgewiesen werden. Somit konnte NadA als erstes Adhäsin der Oca-Proteinfamilie identifiziert werden, das durch Interaktion mit β1 und β3 Integrinen ein Internalisierungssignal induziert. Folglich fungieren β1 und β3 Integrine vermutlich als Wirtszellrezeptoren für nadA-exprimierende Meningokokken. The analysis of the Neisseria meningitidis adhesin NadA in meningococci is hampered due to the presence of diverse neisserial adhesins. Therefore we used Y. enterocolitica as toolbox carrier to characterize NadA functionally and revealed novel insights into the interaction of NadA with epithelial and immune cells, with factors of the complement system and into the role of NadA in pathogenesis in the mouse infection model. Additionally, NadA was found to interact specifically with human β1 and β3 integrins. Thus, NadA is the first Oca-family member which is directly recognized by β1 and β3 integrins and triggers an internalization signal. Therefore β1 and β3 integrins might function as host cell receptors for meningococci expressing nadA.
