Abstract

Amaç: İn siliko analiz kullanılarak Renal Hücreli Karsinom (RHK) patogenezinde rol alabileceği öngörülen potansiyel ceRNA aktivitesi gösterebilecek genlerin belirlenmesi amaçlandı. Daha sonra karşılaştırmalı olarak RHK'li hastalardan alınan tümör ve tümörün çevresindeki normal renal biyopsi örneklerinde bu genlerin ekspresyon düzeylerini incelemeyi ve klinik parametrelerle ilişkilendirmeyi hedefledik. Materyal ve Metot: İn siliko analiz sonucunda 3. kromozomda RHK'ye özgü delesyona uğrayan genlerden yola çıkarak dört muhtemel ceRNA (ATXN3, ABI2, GOLGB1 ve SMAD2) belirlendi. Ondokuz RHK'li hastanın tümör dokusu ile aynı hastanın tümör çevresindeki sağlıklı böbrek dokusunda bu genlerin ifade düzeyleri eş zamanlı PCR ile ölçüldü. Dört genin ifade düzeyi tümör ve çevresindeki normal böbrek dokusu arasında kıyaslandı ve sonra hastaların klinik parametreleriyle ilişkilendirildi. Bulgular: Çalışmamızın sonucunda, tümör dokularında normal böbrek dokularına göre ATXN3 ve GOLGB1 ifade düzeyleri yüksek, ABI2 ifade düzeyi ise düşük bulunmuştur (sırasıyla p=0,936, p=0,546, p=0,494). SMAD2 geninin tespit edilemeyecek kadar az ifade edildiği gözlemlendi. Hastaların ATXN3, ABI2 ve GOLGB1 gen ifade düzeyi değişimleri yaş, cinsiyet, tümör nod metastaz (TNM) evreleri ve Fuhrman grade parametreleri ile ilişkilendirildiğinde anlamlı bir ilişki bulunamadı. ATXN3 geninin direkt bilirubin, monosit eozinofil, bazofil, PCT ve LUC değerleri ile, ABI2 geninin sodyum, klor, glukoz, direk bilirubin, MCV, PLT, eozinofil, bazofil, PCT, PDW, LUC, % PT ve PLT/lenfosit oranı ile ve GOLGB1 geninin ise total bilirubin, direkt bilirubin, monosit eozinofil, bazofil, PCT ve LUC değerleri ile anlamlı derecede ilişkili olduğu belirlendi. Sonuç: İn siliko ceRNA analizi yaklaşımı ve gen ifadesi ölçümü, RHK ile ilişkisi daha önce kanıtlanmış genlerden yola çıkarak yeni hedef genlerin (ATXN3, ABI2 ve GOLGB1) belirlenmesinde akılcı ve ekonomik bir yaklaşım olabilir.Anahtar Kelimeler: ceRNA; Epigenetik; miRNA; Onkogen; Renal hücreli karsinom

Aim: We aimed to determine the genes that could predict the potential ceRNA activity and could be involved in the pathogenesis of RCC in silico analysis. We then aimed to analyze the expression levels of these genes in tumor and their surrounding healthy kidney tissues of patients with RCC and correlate them with clinical patameters. Material and Method: Via in silico analysis upon RCC-specific deleted genes in chromosome 3, four possible ceRNAs (ATXN3, ABI2, GOLGB1 and SMAD2) were identified, expression levels of these genes in tumor tissues of 19 RCC patients and healthy kidney tissues surrounding tumors of the same patients were determined by Real Time PCR, the expression levels of these genes were firstly compared between tumor and healthy kidney tissues and eventually they were correlated with the clinical parameters of the patients.Results: As a result of our study, the expression levels of ATXN3 and GOLGB1 were higher and the expression level of ABI2 was lower in tumor tissues compared to healthy tissues (p=0,936, p=0,546, p=0,494). It has been observed that SMAD2 gene was expressed too low to detect. No significant association was found when patients' ATXN3, ABI2 and GOLGB1 gene expression level changes were associated with TNM stages, Fuhrman grade, age and gender parameters. The ATXN3 gene was significantly associated with direct bilirubin, monocyte eosinophil, basophil, PCT and LUC values, ABI2 gene was significantly correlated with sodium, chlorine, glucose, direct bilirubin, MCV, PLT, eosinophil, basophil, PCT, PDW, LUC,% PT and PLT/lymphocyte ratio and GOLGB1 gene was found to be significantly related to total bilirubin, direct bilirubin, monocyte eosinophil, basophil, PCT and LUC values.Conclusion: In silico ceRNA analysis approach and measurement of gene expression has been a rational and economic approach to the identification of new target genes (ATXN3, ABI2 and GOLGB1) from previously proven genes associated with RCC.Keywords: ceRNA; Epigenetics; miRNA; Oncogene; Renal cell carcinoma
