Studies on the Epizootiological Significance of some Species of Mycoplasma Isolated from Nasal Swabs and Lungs of Pigs

Abstract

SummaryIn a herd free from enzootic pneumonia (Su) a total of 12 Mycoplasma strains were isolated from 200 nasal swabs and 101 macroscopically unchanged lungs. Using the growth‐inhibition test, eleven of them were characterized as Mycoplasma granularum and one as M. laidlawii.In another herd (Tr), infected with enzootic pneumonia, all the 61 nasal swabs were Mycoplasma‐positive; fifty‐seven of them were characterized serologically. Fifty‐six of them were identified as M. hyorhinis and one was not related to any of the type strains (M. hyorhinis, M. granularum, B 3 and B 4). From 33 lungs examined, in the great majority of which macroscopic and microscopic findings corresponded to those found in enzootic pneumonia, 27 strains of Mycoplasma were isolated. Of these isolates, 15 were proved serologically to be M. hyorhinis. Besides small colonies of M. hyorhinis, some isolates showed single large colonies which, after cloning, proved not to be serologically related to any of the prototype strains.The metabolism‐inhibition test carried out with three serologically distinct Mycoplasma strains (M. hyorhinis — Tr 32, M. granularum — Su 24, and a non‐identified strain of Tr 26) revealed low antibody levels (1 : 2 to 1 : 8) against M. hyorhinis in 85 per cent of 62 sera tested in herd Tr and none in herd Su.ZusammenfassungUntersuchungen über die epidemiologische Bedeutung einiger Mycoplasma‐Arten, die aus Nasenabstrichen und Lungen von Schweinen isoliert wurdenBei einem von enzootischer Pneumonie freien Bestand (SU) wurden aus 200 Nasenabstrichen und 101 makroskopisch unveränderten Lungen insgesamt 12 Mycoplasma‐Stämme isoliert. Mittels des Wachstumshemmungstestes wurden 11 von ihnen als M. granularum und einer als M. laidlawii identifiziert.In einem anderen Bestand (Tr) mit enzootischer Pneumonie waren sämtliche 61 entnommenen Nasenabstriche Mycoplasma‐positiv; 57 Stämme wurden serologisch identifiziert; 56 erwiesen sich als M. hyorhinis, ein weiterer hatte zu den Typstämmen keine Verwandtschaft (M. hyorhinis, M. granularum, B 3 und B 4). Aus 33 untersuchten Lungen, die größtenteils makro‐ und mikroskopisch die Veränderungen der enzootischen Pneumonie zeigten, ließen sich 27 Mycoplasma‐Stämme isolieren. Davon erwiesen sich 15 serologisch als M. hyorhinis. Neben den kleinen M. hyorhinis‐Kolonien fanden sich in einzelnen Isolaten auch größere Kolonien, bei denen sich nach Reinzüchtung keinerlei serologische Verwandtschaft mit irgendeinem der Typstämme nachweisen ließ.Der Stoffwechsel‐Hemmtest, der mit drei serologisch verschiedenen Myco‐plasma‐Stämmen ausgeführt wurde (M. hyorhinis‐Stamm Tr 32, M. granularum — Su 24 und einem nicht identifizierten Stamm — Tr 26) ergab im Bestand Tr bei 53 (= 85%) von 62 getesteten Seren niedrige Antikörpertiter (1 : 2 bis 1 : 8) gegen M. hyorhinis, im Bestand Su jedoch nicht.RésuméDans une exploitation exempte de pneumonie enzootique (Su), on isole 12 souches de mycoplasmes à partir de 200 frottis nasaux et de 101 poumons sans modifications macroscopiques. A l'aide du test de l'inhibition de la croissance, on identifie 11 d'entre elles comme M. granularum et une comme M. laidlawii.Dans une autre exploitation atteinte de pneumonie enzootique (Tr), on trouve 61 des frottis nasaux prélevés, positifs aux mycoplasmes; 57 souches furent identifiées sérologiquement; 56 s'avèrent être M. hyorhinis, une autre ne s'apparente à aucune des souches types (M. hyorhinis, M. granularum, D 3 et D 4). Sur 33 poumons examinés, montrant pour la plupart les modifications macro‐ et microscopiques de la pneumonie enzootique, on isole 27 souches de mycoplasmes. 15 d'entre elles s'avèrent sérologiquement identi‐qeus à M. hyorhinis. A côté des petites colonies de M. hyorhinis, on trouve dans quelques cas des colonies plus grandes, qui, au repiquage, ne manifestent aucune parenté sérologique avec les souches types.Le test d'inhibition de métabolisme, pratiqué sur trois souches de mycoplasmes sérologiquement différentes (M. hyorhinis — Tr 32, M. granularum — Su 24 et une souche non identifiée — Tr 26) a donné des titres d'anticorps bas (1 : 2 à 1 : 8) dans l'exploitation Tr, chez 53 sur 62 des sérums testés (85%), mais non dans l'exploitation Su.ResumenEstudios sobre la importancia epidemiológica de algunas especies Mycoplasma, aisladas a partir de raspados nasales y de pulmones porcinosEn una porqueriza (Su), libre de neumonía enzooótica, se aislaron 12 estirpes Mycoplasma partiendo de 200 raspados nasales y 101 pulmones no alterados macroscópicamente. Con ayuda de la prueba de inhibición del crecimiento se identificaron 11 de ellas como M. granularum y una como M. laidlawii.En otra explotación (Tr), con neumonía enzoótica, todas las 61 muestras nasales raspadas eran mycoplasma‐positivas; 57 estirpes se reconocieron sero‐lógicamente; 56 resultaron ser M. hyorhinis, otra no estaba emparentada con las estirpes tipo (M. hyorhinis, M. granularum, D 3 y D 4). A partir de 33 pulmones examinados, que mostraban en su mayor parte las alteraciones macro y microscópicas de la neumonía enzoótica, se lograron aislar 27 cepas Mycoplasma. 15 de éstas resultaron ser M. hyorhinis. Al lado de las colonias diminutas de M. hyorhinis también se hallaron en aislamientos espontáneos unas colonias mayores, en las cuales tras su cultivo puro no se pudo apreciar ningún parentesco serológico con cualquiera de las estirpes tipo.La prueba de inhibición del metabolismo, efectuada con tres cepas Mycoplasma serológicamente diferentes (M. hyorhinis — Tr 32, M. granularum — Su 24 y una estirpe no identificada — Tr 26), tuvo por resultado en la explotación Tr en 53 (= 85%) de 62 sueros sanguíneos analizados unos títulos bajos de anticuerpos (1 : 2 hasta 1 : 8) frente a M. hyorhinis, mientras que no en el efectivo Su.
