Überleben, Differenzierung und funktionelle Auswirkungen transplantierter CSM14.1 Progenitorzellen in neonatalen 6-OHDA-läsionierten Ratten

Abstract

The present study analyzed whether grafts of the mesencephalic progenitor cell line CSM14.1 into the neonatal rat CPu differentiate into neurons and whether this is accompanied by a functional improvement in 6-OHDA-lesioned animals. Rats bilaterally lesioned at postnatal day 1 by intraventricular 6-OHDA-injections 2 days later received 100000 CSM14.1 cells into the right CPu. Five weeks after grafting, the cylinder test was performed. Brain slices immunostained for tyrosine hydroxylase (TH) were quantified by optical densitometry. CSM14.1 cells differentiate into TH-containing neurons, which contribute to the preferred forelimb use, indicating a functional integration of CSM14.1 cells.

Wir untersuchten ob sich mesenzephale Progenitorzellen der Linie CSM14.1 in Hirnen neonatal 6-OHDA läsionierter Ratten neuronal differenzieren und ob dies mit einer Verbesserung motorischer Leistungen einhergeht. Die Tiere wurden am Tag 1 durch intraventrikuläre 6-OHDA Injektion bilateral läsioniert und erhielten 2 Tage später ca. 100.000 CSM14.1 Zellen in das rechte CPu. 5 Wochen danach durchliefen sie einen Zylindertest und anschließend wurden die Hirne immunhistochemisch aufgearbeitet. Die TH-Färbungen wurden densitometrisch quantifiziert. Es zeigte sich eine Differenzierung der CSM 14.1 Zellen in TH-positive Neurone, die mit einer motorischen Verbesserung der Tiere einherging.
