Combined Expression of Metastasis Related Markers Naa10p, SNCG and PRL-3 and its Prognostic Value in Breast Cancer Patients

Abstract

Les combinaisons de plusieurs biomarqueurs représentant des aspects distincts de la métastase peuvent avoir une meilleure valeur pronostique pour les patientes atteintes d'un cancer du sein, en particulier celles qui en sont à un stade avancé. Dans cette étude, nous avons évalué les niveaux protéiques de la protéine N-α-acétyltransférase 10 (Naa10p), de la synucléine-γ (SNCG) et de la phosphatase du foie 3 régénérant (PRL-3) chez 365 patientes atteintes d'un cancer du sein par immunohistochimie. Des sous-groupes pronostiques distincts du cancer du sein ont été identifiés par la combinaison des trois biomarqueurs. Le sous-groupe Naa10p+ SNCG-PRL-3 a montré le meilleur pronostic avec une survie sans métastase (DMFS) médiane à distance de 140 mois, tandis que le sous-groupe Naa10p-SNCG +PRL-3+ avait le pire pronostic avec une DMFS médiane de 60,5 mois. L'analyse multivariée a indiqué que Naa10p, SNCG, PRL-3 et la classification TNM étaient tous des facteurs pronostiques indépendants pour la DMFS et la survie globale (OS). La combinaison des trois biomarqueurs Naa10p, SNCG et PRL-3 a mieux fonctionné chez les patients atteints de métastases ganglionnaires, en particulier ceux présentant des tumeurs plus avancées que les autres sous-groupes. En conclusion, le profil d'expression combiné de Naa10p, SNCG et PRL-3, seul ou en combinaison avec le système de classification TNM, peut fournir une estimation précise du pronostic des patients atteints de cancer du sein.

Las combinaciones de múltiples biomarcadores que representan distintos aspectos de la metástasis pueden tener un mejor valor pronóstico para los pacientes con cáncer de mama, especialmente aquellos en etapas tardías. En este estudio, evaluamos los niveles de proteína de la proteína N-α-acetiltransferasa 10 (Naa10p), sinucleína-γ (SNCG) y fosfatasa de regeneración hepática-3 (PRL-3) en 365 pacientes con cáncer de mama mediante inmunohistoquímica. Se identificaron distintos subgrupos de pronóstico de cáncer de mama mediante la combinación de los tres biomarcadores. El subgrupo Naa10p+ SNCG-PRL-3 mostró el mejor pronóstico con una mediana de supervivencia libre de metástasis a distancia (DMFS) de 140 meses, mientras que el subgrupo Naa10p-SNCG +PRL-3+ tuvo el peor pronóstico con una mediana de DMFS de 60,5 meses. El análisis multivariante indicó que Naa10p, SNCG, PRL-3 y la clasificación TNM fueron todos factores de pronóstico independientes tanto para DMFS como para la supervivencia general (SG). La combinación de tres biomarcadores de Naa10p, SNCG y PRL-3 tuvo un mejor desempeño en pacientes con metástasis en los ganglios linfáticos, especialmente aquellos con tumores más avanzados que otros subgrupos. En conclusión, el perfil de expresión combinado de Naa10p, SNCG y PRL-3, solo o en combinación con el sistema de clasificación TNM, puede proporcionar una estimación precisa del pronóstico de los pacientes con cáncer de mama.

Combinations of multiple biomarkers representing distinct aspects of metastasis may have better prognostic value for breast cancer patients, especially those in late stages.In this study, we evaluated the protein levels of N-α-acetyltransferase 10 protein (Naa10p), synuclein-γ (SNCG), and phosphatase of regenerating liver-3 (PRL-3) in 365 patients with breast cancer by immunohistochemistry. Distinct prognostic subgroups of breast cancer were identified by combination of the three biomarkers.The Naa10p+SNCG-PRL-3-subgroup showed best prognosis with a median distant metastasis-free survival (DMFS) of 140 months, while the Naa10p-SNCG+PRL-3+ subgroup had the worst prognosis with a median DMFS of 60.5 months.Multivariate analysis indicated Naa10p, SNCG, PRL-3, and the TNM classification were all independent prognostic factors for both DMFS and overall survival (OS).The three biomarker combination of Naa10p, SNCG and PRL-3 performed better in patients with lymph node metastasis, especially those with more advanced tumors than other subgroups.In conclusion, the combined expression profile of Naa10p, SNCG and PRL-3, alone or in combination with the TNM classification system, may provide a precise estimate of prognosis of breast cancer patients.
