Secuenciación de ADN antiguo de restos humanos del País Vasco

Abstract

Se ha recuperado DNA mitocondrial (región 1 del D-loop) de tres individuos procedentes del yacimiento vasco de época altomedieval de Alto de la Ermita (Amézaga, Araba), mediante la técnica de PCR (reacción en cadena de la polimerasa). Los dientes se han mostrado como un excelente sistema de protección del material hereditario. El estudio del DNA de poblaciones antiguas presenta numerosos problemas técnicos debido al alto grado de degradación del DNA, la inhibición de la amplificación y la posibilidad de contaminación de las muestras. Sin embargo, el DNA antiguo proporciona información genética directa de las poblaciones del pasado, y puede ser útil para establecer relaciones de afinidad de estas poblaciones con los grupos humanos actuales. Hasta el momento, se han obtenido las secuencias de una zona de DNA de 228 bp, correspondientes a dos de los individuos estudiados. La información que proporcionan estas secuencias, sin embargo, no podrá ser valorada adecuadamente hasta que no se disponga de más datos de DNA de poblaciones vascas actuales y pasadas.
