Abstract

[Resumen] LA RETINOSIS PIGMENTARIA ES UNA ENFERMEDAD HEREDITARIA QUE CURSA CON DEGENERACION PROGRESIVA DE LA RETINA Y ES UNA DE LAS CAUSAS MAS FRECUENTES DE PERDIDA DE VISION. EL PATRON DE HERENCIA DE ESTA ENFERMEDAD ES COMPLEJO PORQUE PUEDE SEGREGAR DE MANERA AUTOSOMICA DOMINANTE, RECESIVA Y LIGADA AL CROMOSOMA X. LA RETINOSIS PIGMENTARIA PRESENTA UNA HETEROGENEIDAD GENETICA YA QUE EXISTE MAS DE UN LOCUS PARA CADA UNO DE LOS TIPOS DE HERENCIA MENCIONADOS. SE CONSIDERAN GENES CANDIDATOS AQUELLOS QUE CODIFICAN PARA PROTEINAS QUE DE MANERA DIRECTA O INDIRECTA INTERVIENEN EN LA CASCADA DE TRANSDUCCION VISUAL. EN ESTE TRABAJO SE HAN ANALIZADO DOS GENES, LA SUBUNIDAD BETA DE LA FOSFODIESTERASA (PDEBETA) Y LA PROTEINA DE UNION AL RETINOIDE INTERSTICIAL (IRBP) EN 45 FAMILIAS ESPAÑOLAS AFECTADAS DE RETINOSIS PIGMENTARIA AUTOSOMICA RECESIVA. LOS ESTUDIOS MEDIANTE MARCADORES POLIMORFICOS DE HOMOCIGOSIDAD Y SEGREGACION DE UN HAPLOTIPO CONCRETO CON LA ENFERMEDAD NOS HAN PERMITIDO SELECCIONAR AQUELLAS FAMILIAS EN LAS QUE LOS GENES PROPUESTOS PUDIESEN ESTAR INVOLUCRADOS. COMO RESULTADO DEL ANALISIS DE POLIMORFISMOS DE CONFORMACION DE HEBRA SIMPLE Y SECUENCIACION, SE DESCRIBEN TRES MUTACIONES EN EL GEN PDEBETA EN HOMOCIGOSIS EN TRES FAMILIAS CONSANGUINEAS COMO RESPONSABLES DE LA PATOLOGIA.
