Abstract

[RESUMEN]: La osteoartritis (OA), o enfermedad degenerativa articular, es común en individuos mayores de 65 años, siendo una causa importante de incapacidad laboral. Una de las articulaciones más afectadas es la rodilla. Este estudio analizó la dinámica de pérdida de secuencias teloméricas en leucocitos de sangre periférica tras 6 años de seguimiento y el haplogrupo de ADN mitocondrial (ADNmt), como posibles factores de riesgo para el padecimiento en el momento de reclutamiento (prevalencia) y/o aparición durante el seguimiento (incidencia) de OA de rodilla, evaluada radiológicamente. Los individuos estudiados pertenecen al consorcio Osteoarthritis Iniciative (OAI) de EEUU. La cantidad de secuencias teloméricas fue determinada mediante una qPCR específica y el haplogrupo de ADNmt mediante PCR-RFLP con el método de extensión de una sola base. Se demostró que el porcentaje relativo de pérdida de secuencias teloméricas en leucocitos es un factor de riesgo independiente asociado con la aparición de la OA de rodilla. Los portadores del clúster de haplogrupo de ADNmt HV evidenciaron una mayor incidencia que los portadores del clúster JT, al igual que una mayor pérdida de secuencias teloméricas. El mayor acortamiento telomérico en leucocitos de individuos incidentes podría ser indicativo de un envejecimiento sistémico acelerado que potenciaría la degeneración del cartílago.

[RESUMO]: A artrose (OA), ou enfermidade articular dexenerativa, é común en persoas maiores de 65 anos, sendo unha causa importante de incapacidade laboral. Unha das articulacións máis afectadas é o xeonllo. Este estudo analizou a dinámica de perda das secuencias teloméricas en leucocitos do sangue periférico despois de 6 anos de seguimento e o haplogrupo do ADN mitocondrial (ADNmt), como posibles factores de risco para a condición no momento do recrutamento (prevalencia) e/ou do inicio durante o seguimento (incidencia) da OA de xeonllo, avaliada radiolóxicamente. Os individuos estudados pertencen ao consorcio Osteoartritis Initiative (OAI) dos EEUU. O número de secuencias teloméricas determinouse mediante qPCR específica e o haplogrupo do ADNmt mediante PCR-RFLP co método de extensión dunha única base. A porcentaxe relativa de perda das secuencias teloméricas nos leucocitos demostrou ser un factor de risco independente asociado ao desenvolvemento da OA de xeonllo. Os portadores do clúster de haplogrupo de mtDNA HV mostraron unha maior incidencia que os portadores do clúster JT, así como unha maior perda das secuencias teloméricas. O maior acurtamento dos telómeros nos leucocitos dos individuos incidentes podería ser indicativo dun envellecemento sistémico acelerado que potenciaría a dexeneración da cartilaxe.

[ABSTRACT]: Osteoarthritis (OA) is a degenerative joint disease, which is very common between people aged >65 years old. It is also one of the major causes of occupational disability. Knee is one of the most affected joints. In this study, we analyzed the dynamics of telomeric DNA sequence loss in peripheral blood leukocytes (PBL) after a 6 years follow-up and the mitochondrial DNA haplogroup (mtDNA), as possible risk factors for suffering knee OA at the recruitment (prevalence) or for developing it during follow-up (incidence). The diagnosis was performed by radiologic images. The subjects studied belong to the Osteoarthritis initiative (OAI) from EEUU. The amount of telomeric sequences was determined by specific qPCR whereas mtDNA haplogroup was determined by PCR-RFLP using the single base extension method. It was evidenced that the relative percentage of telomeric sequences loss in PBL is an independent risk factor associated with knee OA onset. Carriers of mtDNA HV haplogroup clúster showed a higher incidence of OA and telomeric sequence loss than carriers of JT cluster. The higher telomere shortening found in leukocytes from patients who develop OA could be indicative of an accelerated systemic aging process that would potentiate cartilage degeneration.
