Abstract

[Resumen] Se realizó el microinjerto in vitro de la variedad ‘Branca’ de castaño (Castanea sativa Mill.) sobre los portainjertos híbridos (Castanea sativa x C. crenata) 3 y 7521, con el objetivo de analizar diversas variables que pueden influir en el éxito del microinjerto en castaño para la producción de planta. Se analizó el efecto del genotipo del portainjerto, del protocolo de enraizamiento previo de los portainjertos híbridos, de la presencia o no de raíces en el portainjerto y del tipo de púa (una o dos yemas axilares) sobre la tasa de prendimiento del injerto y el crecimiento de la púa. En los ensayos previos de enraizamiento de los portainjertos se evaluaron las tasas de enraizamiento y de necrosis apical de microbrotes con o sin yema apical y defoliados o sin defoliar, con el objetivo de analizar otros factores que pueden influir de forma directa o indirecta en la producción de planta de calidad. Se demostró que la eliminación de la yema apical en los portainjertos disminuye el porcentaje de plantas necrosadas durante el proceso de enraizamiento y que la defoliación de los portainjertos conduce a menores porcentajes de enraizamiento y de prendimiento del microinjerto. Asimismo, la presencia o no de raíces no influye en la tasa de prendimiento ni en el crecimiento de la púa tras el microinjerto, y el uso de púas con dos yemas mejora la tasa de prendimiento

[Resumo] O microenxerto in vitro da variedade de castiñeiro 'Branca' (Castanea sativa Mill.) realizouse sobre os portaenxertos híbridos (Castanea sativa x C. crenata) 3 e 7521, co obxectivo de analizar diversas variables que poden influír no éxito do microenxerto en castiñeiro para produción vexetal. Analizouse o efecto do xenotipo do portaenxerto, o protocolo de enraizamento previo dos portaenxertos híbridos, a presenza ou ausencia de raíces no portaenxerto e o tipo de puga (un ou dous xemas axilares) sobre a taxa de toma do enxerto e o crecemento da puga. Nos anteriores ensaios de enraizamento dos portaenxertos avaliáronse as taxas de enraizamento e necrose apical dos microbrotes con ou sen xema apical e defoliados ou sen follas, co obxectivo de analizar outros factores que poidan influír directa ou indirectamente na produción de planta de calidade. Demostrouse que a eliminación da xema apical nos portaenxertos diminúe a porcentaxe de plantas necrosadas durante o proceso de enraizamento e que a defoliación dos portaenxertos conduce a menores porcentaxes de enraizamento e toma do microenxerto. Así mesmo, a presenza ou ausencia de raíces non inflúe na taxa de adherencia nin no crecemento da espiga despois do microenxerto, e o uso de espigas con dúas xemas mellora a taxa de adherencia.

[Abstract] The in vitro micrograft of the chestnut variety 'Branca' (Castanea sativa Mill.) was carried out on the hybrid rootstocks (Castanea sativa x C. crenata) 3 and 7521, with the aim of analyzing various factors that can influence the success of the micrograft in chestnut for plant production. The rootstock genotype, previous rooting protocol of the hybrid rootstocks, presence or absence of roots in the rootstock and type of scion (with one or two axillary buds) effect on the graft take rate and the scion growth was analyzed. In the previous rooting trials of the rootstocks, the rooting and apical necrosis rates of microshoots with or without apical bud, and defoliated or undefoliated were evaluated, with the aim of analyzing other factors that may directly or indirectly influence production of quality plants. It was shown that the elimination of the apical bud in the rootstocks decreases the percentage of necrotic plants during the rooting process and that the defoliation of the rootstocks leads to lower percentages of rooting and taking of the micrograft. On the other hand, the presence or absence of roots does not influence the graft-take rate or the growth of the scion after micrografting, and the use of scions with two buds improves the graft-take rate.
