Abstract

[Resumen]: La artrosis (OA) es una patología que afecta al cartílago articular, provocando su degeneración. Recientemente se ha destacado la importancia de los procesos inflamatorios y de la regulación epigenética sobre su progresión, y también se ha asociado una mayor adherencia a la dieta mediterránea con una menor prevalencia de OA. Teniendo en cuenta el carácter crónico y la falta de un tratamiento efectivo contra esta enfermedad, el presente trabajo constituye un estudio del efecto de nutracéuticos extraídos del aceite de oliva virgen extra (oleocantal y ligustrósido de aglicón) sobre tejido normal y artrósico a nivel de expresión génica y de liberación de óxido nítrico, gas de gran relevancia en la patogénesis de la OA. Además, incluye un análisis histológico de la matriz del cartílago, de las marcas epigenéticas 5hidroximetilcitosina y 5-metilcitosina, y de la modificación H3K27me3; todas ellas implicadas en la OA.

[Abstract]: Osteoarthritis (OA) is a disease that affects the articular cartilage, causing its degeneration. Recently, the importance of inflammatory processes and epigenetic regulation on its progression has been highlighted, and greater adherence to the Mediterranean diet has also been associated with a lower prevalence of OA. Taking the chronic nature and the lack of an effective treatment against this disease into account, the present work constitutes a study of the effect of nutraceuticals extracted from extra virgin olive oil (oleocanthal and ligstroside aglycone) on normal and arthritic tissue at the level of gene expression and release of nitric oxide, a gas of great relevance in the OA pathogenesis. In addition, it includes a histological analysis of the cartilage matrix, the epigenetic marks 5-hydroxymethylcytosine and 5-methylcytosine, and the H3K27me3 modification; all of which are implicated in OA.

[Resumo]: A artrose (OA) é unha patoloxía que afecta á cartilaxe articular, provocando a súa dexeneración. Recentemente destacouse a importancia dos procesos inflamatorios e da regulación epixenética na súa progresión, e tamén asociouse unha maior adherencia á dieta mediterránea cunha menor prevalencia de OA. Tendo en conta o carácter crónico e a falta dun tratamento efectivo contra esta enfermidade, o presente traballo constitúe un estudo do efecto de nutracéuticos extraídos do aceite de oliva virxe extra (oleocantal e ligustrósido de aglicón) sobre tecido normal e artrósico a nivel de expresión xénica e de liberación de óxido nítrico, gas de gran relevancia na patoxénese da OA. Ademáis, inclúe unha análise histolóxica da matriz da cartilaxe, das marcas epixenéticas 5-hidroximetilcitosina e 5-metilcitosina, e da modificación H3K27me3; todas elas implicadas na OA.
