Abstract

[Abstract] Introduction: Tobacco is a worldwide relevance problem. According to the WHO (World Health Organization) tobacco causes the death of more than 8 million people a year. In addition, it has negative effects on both smokers and people who inhale tobacco smoke. Children are also affected by this second-hand smoke, which can cause serious damage, including death. To prevent tobacco from being the cause of disease and death, new drugs, varenicline and bupropion, have been made available to the population to improve the efficacy of the therapies known so far for smoking cessation. Objective: The objective of our study is to identify the real efficacy of varenicline and bupropion for smoking cessation. Methodology: To answer the study question and the stated objectives, a systematic review was carried out. A search for clinical trials was conducted in Pubmed, Scopus and Cochrane. The search strategy was limited to studies in the last 11 years in english or spanish. Results: Both vareniclina and bupropion are more effective than any tobacco cessation therapy used so far. Varenicline (OR 2,15-4,7) has greater efficacy than bupropion (OR 0,86-2,84), especially in the long term, but some authors argue that this efficacy is not clinically significant. The combination of both therapies (OR 1,09-3,71) improves the effectiveness of their use as monotherapy without increasing adverse events. Also is important to mention the association between determined genes like CYP2B6 and its variant rs8109525 and the efficacy of the treatment. Conclusions: There is an increase in efficacy with the use of these drugs either as monotherapy or combining both drugs. Furthermore, in the treatment with varenicline and bupropion their haven´t been identified serious adverse events. Research must continue in this area to find new ways to improve the effectiveness of these treatments.
