Abstract

[Resumen] La variabilidad espacial y temporal del bacterioplancton está relacionada con diversos factores tanto bióticos como abióticos. En este trabajo se estudió la estructura y composición de las comunidades bacterianas a lo largo de una sección perpendicular a la costa de A Coruña (43°N, 8°W) durante la campaña Hercules III (Julio 2012). Se estudiaron dos zonas de la columna de agua: la superficie (SUP, 0-30 m) y el máximo de clorofila (DCM [depth chlorophyll maximum], 30-100 m), con el objetivo de estudiar su relación con las variables medioambientales medidas en la columna de agua. La abundancia y composición bacteriana se determinó mediante citometría de flujo, hibridación fluorescente in situ (CARD-FISH), y el análisis de pirosecuenciación de la región HTS del 16S rRNA. Proteobacteria fue el filum más abundante a lo largo de la columna de agua, dominando Alphaproteobacteria en superficie y Gammaproteobacteria en el DCM. Mientras que la comunidad de la superficie estuvo compuesta por bacterias de la familia Flavobacteriaceae (filum Bacteriodetes) y el cluster SAR11 (Alphaproteobacteria), en el DCM dominaron la familia Ocenospirillales (Gammaproteobacteria) representada por el cluster SAR86. El análisis de escalamiento multidimensional nMDS mostró una correlación entre las variables ambientales estudiadas y los filos taxonómicos con la estructura de la composición de las comunidades bacterianas, indicando que existen diferencias significativas entre las comunidades bacterianas de superficie y el máximo de clorofila. Mientras que la temperatura y la radiación fotosintéticamente activa (PAR) determinan la diversidad bacteriana de superficie, en el DCM estos cambios estuvieron significativamente correlacionados con la salinidad, la concentración de clorofila a y de nutrientes inorgánicos. Nuestro estudio concluye que existe una clara estratificación vertical en la composición de las comunidades bacterianas, que se correlacionó significativamente con las variables ambientales medidas en la costa de A Coruña.

[Abstract] Spatial and temporal variability of bacterioplankton are related to several biotic and abiotic factors. In this work, the structure and composition of bacterial along a line perpendicular to the coast of A Coruña communities section (43 ° N, 8 ° W) were studied during the Hercules III campaign (July 2012). Two areas of the water column were studied: the surface (SUP, 0-30 m) and chlorophyll maximum (DCM [chlorophyll maximum depth], 30-100 m), in order to study its relation to environmental variables measured in the water column. Bacterial abundance and composition was determined by flow cytometry, fluorescence in situ hybridization (CARDFISH), and pyrosequencing analysis of 16S rRNA region HTS. Proteobacteria was the most abundant phylum throughout the water column, dominating Alphaproteobacteria in the surface and Gammaproteobacteria in DCM. While the surface community was composed of bacteria Flavobacteriaceae family (phylum Bacteriodetes) and SAR11 cluster (Alphaproteobacteria), in DCM dominated Ocenospirillales family (Gammaproteobacteria) represented by the SAR86 cluster. The nMDS multidimensional scaling analysis showed a correlation between environmental variables and taxonomic phyla with the structure of the composition of bacterial communities, indicating that there are significant differences between the surface and chlorophyll maximum bacterial communities. While temperature and photosynthetically active radiation (PAR) determine the surface bacterial diversity, in DCM these changes were significantly correlated with salinity, chlorophyll a and inorganic nutrients. Our study concludes that there is a clear vertical stratification in the composition of bacterial communities, which were significantly correlated with environmental variables measured on the coast of A Coruña.
