Chaperones moleculares : su rol en el proceso de plegamiento y ensamble intracelular de proteínas

Abstract

El plegamiento de las proteínas a su conformación nativa constituye una de las bases para el correcto funcionamiento celular. La eficiencia del proceso es asegurada in vivo gracias a la participación entre otros, de sistemas especializados de proteínas denominados chaperones moleculares. Estos asistentes de plegado interaccionan no covalentemente con polipéptidos total o parcialmente desplegados favoreciendo el plegado productivo por sobre las reacciones no productivas que conducen a la agregación en condiciones fisiológicas y de estrés. Numerosas evidencias experimentales indican que distintos chaperones pueden cooperar en las células. Se ha propuesto que los sistemas de chaperones Hsp70/DnaK y Hsp60/GroE actúan secuencialmente asistiendo el plegamiento de proteínas recién sintetizadas o desnaturalizadas y el importe de éstas a organelas. Sin embargo, la transferencia secuencial parece no ocurrir en todos los casos y por ello se ha propuesto la existencia de redes flexibles y funcionales …
