Evaluación de la linealidad en inmunoensayos

Abstract

Los inmunoensayos cuantitativos deben validar su propiedad de linealidad, es decir, evaluar en qué rango de concentraciones se cumple exactitud y precisión de manera aceptables. Para hacer esta evaluación se usa la recta de regresión lineal obtenida mediante el método de los mínimos cuadrados ordinarios, pero como los datos observados en los inmunoensayos no cumplen el supuesto de homocedasticidad, se han estudiado diversos métodos alternativos para obtener la mejor recta que se ajuste a los datos: (1) transformación de los datos a través de funciones estabilizadoras de la varianza, (2) aplicación de los mínimos cuadrados ponderados y (3) estimadores no paramétricos de una recta, como son los estimadores de Siegel y Theil - Sen. Además se propone un nuevo protocolo de evaluación de la linealidad que se ha implementado con software R y que automáticamente genera un informe de resultados de gran utilidad para los investigadores.
