Abstract

La transtirretina (TTR) es una proteína plasmática, involucrada en el transporte de la tiroxina y del retinol. Su forma wild-type y sus diferentes mutantes están asociados a diferentes amiloidosis, siendo la mutación Y78F la más amiloidogénica. El presente trabajo tenía como principal objetivo la producción y purificación de lotes de hTTR Y78F y la identificación de posibles modificaciones posttraduccionales en el residuo de cisteína. La expresión de la proteína se dio en E. coli a escala biorreactor y las técnicas básicas de purificación incluyeron disrupción celular, precipitación fraccionada de proteínas y cromatografías de intercambio iónico y de gel filtración. Para la identificación de las modificaciones post-traduccionales se usó la técnica de MALDI-TOF. Durante este trabajo, se pudo confirmar que la producción de TTR en biorreactor, incluso sin haber estado optimizado, es mucho más ventajosa que en erlenmeyer, tanto en términos de cantidad de proteína recuperada como en términos de productividad volumétrica. También se observó que, para las condiciones experimentadas, un tiempo de inducción de aproximadamente 4 horas es lo más indicado. Con relación a las modificaciones post-traduccionales se observó que están presentes a lo largo de toda la inducción, que su perfil temporal no varía en un mismo experimento y que son removibles con el agente reductor DTT.
