Estudio de la ecotoxicidad del hierro en células neuronales

Abstract

L’objectiu d’aquest projecte es conèixer la implicació del complex ferritina-ferro en l’estrès oxidatiu iniciat per el glutamat que te lloc quan es produeix un infart cerebral. Utilitzarem un model de cultius de neurones corticals de rata. En primer lloc s’incuben les celules amb glutamat per obtindre cultius en els que les neurones presentin les característiques bioquímiques pròpies que es produeixen d’infart cerebral. Tot seguit s’analitza l’efecte neurotòxic provocat per la ferritina afegida en el medi de cultiu, determinant el seu efecte sobre l’estrès oxidatiu. La mort cerebral es determinada mitjançant el mètode de la Lactato deshidrogenasa y la concentració de ferro mitjançant espectoftometría. Seguint aquest procediment, concloem que el ferro esta implicat en els mecanismes moleculars que causen la mort neuronal.
