Beyond Viral Suppression: Multi-Omics Analyses into HIV Immunological Non-Responders and cART Toxicity

Abstract

Het Hiv-virus verwoest het immuunsysteem. Dankzij medicijnen herstelt het immuunsysteem zich en wordt HIV een chronische ziekte met een goede levensverwachting. Toch is er een kleine groep bij wie het immuunsysteem zich ondanks medicijnen maar beperkt herstelt; immunological non-responders (INR). Dit proefschrift beschrijft dat INR een tekort hebben aan een specifiek eiwit: SKAP1. Dit komt door veranderingen in de structuur van het opbergen van DNA in de cel, de zogenaamde epigenetica. Ook heeft een deel van het immuunsysteem van INR kenmerken van over-activatie en uitputting. Omdat mensen met HIV hun leven lang medicatie moeten gebruiken, is er risico op nadelige effecten op lange termijn. Dit proefschrift beschrijft hoe verschillende soorten medicijnen een verschillende invloed hebben op de energievoorziening van cellen en laat zien dat minder HIV medicatie waarschijnlijk beter is voor het immuunsysteem.
