Cumarinas en la prevención de estres oxidativo dependiente de la edad

Abstract

Este estudio relaciona el comportamiento animal y la concentración de antioxidantes endógenos con el envejecimiento biológico, como consecuencia del tratamiento farmacológico con cumarinas. Hemos revisado el estado actual de los conocimientos sobre: teorías de envejecimiento, radicales libres y peroxidación tisular, antioxidantes, y cumarinas como potenciales agentes antioxidantes. La experimentación se ha iniciado con un screening de 19 cumarinas mediante técnicas in vitro de captación de radicales. A partir de éstas, se han seleccionado las cumarinas con mayor actividad anti-radicalaria. Dichas cumarinas se administraron de forma crónica a ratones c57bl/6j de distintas edades. Durante el tratamiento se ha evaluado el comportamiento (vitalidad, coordinación motora, memoria,...), y al final del mismo, los animales se han sacrificado y se ha extraido hígado y corteza cerebral. En estas muestras se han cuantificado diversos antioxidantes y el grado de daño tisular para conocer la influencia de las cumarinas en la prevención del daño oxidativo dependiente de la edad
