Aproximación a la síntesis del fragmento espiroacetálico de avermectina-milbemicina

Abstract

Se ha estudiado una aproximación convergente a partir de d-glucosa para la síntesis enantioselectiva del fragmento espiroacetalico de avermectinas y milremilinas, producto de importante interés farmaceútico y a su vez de gran complejidad estructural. Siguiendo un planteamiento retrosintético inicial que divide el espiroaletal en dos subestructuras, electrófilo y nucleófilo, se ha desarrollado metodología original que permite obtener cada una de dichas subunidades con la esteroquímica deseada. Así mismo, y para el acoplamiento final se ha desarrollado un nuevo método de formación de espiroacetales insaturados que implica a)alquilación estereoselectiva de moglicosidos insaturados con sulfatos cíclicos, y b)reacción de glicosidación intramolecular en medio neutro
