Efecto del glifosato sobre la simbiosis Lupinus albus-Bradyrhizobium sp. (Lupinus)

Abstract

En esta tesis se lleva a cabo un estudio integrado de la simbiosis Bradyrhizobium - Lupinus albus y del efecto que sobre esta simbiosis produce, el herbicida glifosato. Este herbicida afecta drásticamente a la fijación simbiótica de nitrógeno en plantas de lupino. El descenso de la actividad nitrogenasa se debe a la inhibición de procesos fisiológicos en el citosol del nódulo. La aplicación de glifosato produce cambios en el metabolismo de carbono. El descenso del contenido de almidón paralelamente al de la actividad sacarosa sintasa y el aumento de sacarosa, se relacionan con la alteración de la ruta de biosíntesis de aminoácidos aromáticos por el glifosato. Asimismo, el glifosato produjo cambios en la ultraestructura del nódulo y del aparato fotosíntético, progresivos y diferenciales. Las concentraciones del ácido shikímico y protocatéquico aumentan en los nódulos y hojas de Lupinus albus rápidamente después de la aplicación del herbicida. El tratamiento con glifosato ha puesto de manifiesto una correlación directa entre la acumulación de compuestos fenólicos y el descenso de la actividad nitrogenasa. La detección de un nuevo polipéptido de 44 kDa en el patrón proteico del citosol bacteroidal fue la alteración más destacada producida por el glifosato en el metabolismo de proteínas. Esta proteína (BLpp) está codificada por un único gen en el genoma bacteriano y estudios de la expresión génica de esta proteína indican que los niveles de expresión del gen blpp aumentan en los nódulos a medida que aumenta la concentración del glifosato. Asimismo en la estructura de BLpp se han identificado un posible motivo de asociación a membrana y un dominio característico de la familia de porinas, lo que sugiere que BLpp es una posible porina. Esta sería la primera porina descrita en la bacteria Bradyrhizobium sp. (Lupinus).
