Abstract

Mediante este trabajo de tesis hemos demostrado que el Factor de Crecimiento tipo Insulina 1 (el IGF-I) tiene, al margen de sus funciones clasicas como factor trofico, efectos moduladores de la actividad de los receptores ionotrópicos de glutamato. Empleando tecnicas electrofisiológicas hemos podido comprobar que el IGF-I es capaz de modular de forma diferencial la actividad de los distintos tipos de receptores ionotropicos de glutamato presentes en las celulas granulares de cerebelo. Esta modulacion es rapida(inferior a 3 minutos) y especifica para cada tipo de receptor y tendria una enorme importancia funcional debido al gran numero de procesos en los que participan estos receptores entre los que se incluyen procesos de aprendizaje y memoria en el individuo adulto. Tambien en este trabajo de tesis hemos identificado toda una serie de nuevas proteinas que se asocian al receptor de AMPA y que son susceptibles de ser moduladas por efecto del IGF-I. Asimismo hemos descrito un posible mecanismo mediador del efecto del IGF-I a traves de un proceso de internalizacion de algunas subunidades de los receptores de AMPA. Según nuestro conocimiento, esta es la primera observacion de un efecto regulador de la función de un receptor por modulacion simultanea de numerosos eventos (internalización, fosforilacion/desfosforilacion y asociación/disolucion de proteinas) por la influencia de un unico modulador: el IGF-
