Estandarización de una técnica de complementación del gen pncA en Mycobacterium tuberculosis pncA-knockout: herramienta para el estudio de la relación entre mutaciones en pncA y parámetros fenotípicos

Abstract

Mutaciones en el gen pncA son la mayor causa de resistencia a pirazinamida (PZA), pero existen mecanismos alternativos que también aportan resistencia. El objetivo del estudio fue estandarizar la técnica de complementación del gen pncA en Mycobacterium tuberculosis H37Rv pncA-KO para evaluar el efecto de las mutaciones en el gen pncA sobre parámetros fenótipicos, en un escenario controlado por la variabilidad genética de los aislados clínicos de donde provienen los genes pncA mutantes. Se generaron cepas complementadas con genes pncA WT y mutantes D49N, H51R, G78C y F94L a través del sistema de expresión pNIT-1 para evaluar la actividad PZAsa mediante la prueba de Wayne cuantitativa, nivel de expresión del gen pncA y susceptibilidad a PZA usando concentraciones graduales de PZA en condiciones no inducida e inducida. El sistema de expresión pNIT-pncA restauró el fenotipo de la cepa pncA-KO y los plásmidos se mantuvieron estables intracelularmente durante el período de observación que duró 18 semanas. Los niveles de expresión basal resultaron constantes y similares al de la cepa H37Rv. La cepa pncA WT complementada mostró actividad PZAsa y Concentración Mínima Inhibitoria de PZA similar al de la cepa H37Rv. Sin embargo, las cepas pncA mutantes complementadas fueron resistentes a PZA en ambas condiciones y la sobreexpresión de los genes pncA mutantes permitió verificar la pérdida total o parcial de la actividad PZAsa.
