Análisis transcriptómico de la respuesta a heladas en papas nativas

Abstract

Las heladas constituyen un fenómeno recurrente que afectan la producción y el rendimiento de los cultivos como la papa. En el Perú existen algunas especies de papas nativas tolerantes a heladas que constituyen un importante reservorio genético para el fitomejoramiento de este tubérculo. En esta investigación se caracterizó el transcriptoma de la papa en respuesta a heladas utilizando la técnica RNAseq. Se trabajó con una especie de papa nativa tolerante (Solanum goniocalyx) y una susceptible (S. stenotomum). Se generaron 24 bibliotecas con 31 millones de lecturas por biblioteca. Se observó que aproximadamente 80% de lecturas eran de mapeo único, 9% de mapeo múltiple y 12% no mapean al genoma de referencia. Comparando los niveles de expresión, se notó que la especie tolerante expresaba diferencialmente entre 354 y 629 genes, mientras que la susceptible, entre 1,590 y 3,164. Adicionalmente, se notó que, en la especie tolerante 110 genes son específicamente inducidos durante la respuesta temprana al estrés. Finalmente, se logró correlacionar los perfiles de expresión diferencial entre los genes, obteniéndose 7 módulos (por especie). Esta investigación permitió generar información importante para estudios posteriores en fitomejoramiento de papas de importancia alimenticia y económica que se encuentren perjudicadas por las recurrentes bajas temperaturas.
