Abstract

Çalışmamızda Holstein ırkı sığıra ait göz organının mikroskobik yöntemler kullanılarak morfolojik ve morfometrik özelliklerinin ortaya konması amaçlandı. Bu amaç doğrultusunda, 20 adet sığıra ait sağ ve sol toplamda 40 göz, etrafındaki dokulara zarar vermeden orbita'dan çıkarıldı. Stereomikroskop altında gözlerin morfolojik görüntüleri fotoğraflandı. Fiksasyon aşamasına geçilmeden önce external morfometrik ölçümler elde edildi. Diseksiyon sonrası tespit edilen her bir gözden hem elektron hem de ışık mikroskopta incelemek üzere ayrı kesitler alındı. Işık mikroskobik incelemelerin yapılması için tespit sonrası doku örnekleri parafine gömülerek 5 mikrometre kalınlıkta alınan kesitlere Crossmann'ın üçlü boyama tekniği uygulandı.Tunica fibrosa bulbi'nin anterior 1/6'sını cornea'nın geri kalan kısmı ise sclera'nın oluşturduğu görüldü. Cornea'nın anterior yüzünü kaplayan epithelium anterius cornea çok katlı yassı non-keratinize epitelden oluşmaktaydı. Elektron mikroskop altında cornea'nın substantia propria katmanının içerdiği yoğun kollajen liflerden dolayı dalgalı bir görüntü verdiği gözlendi. Işık mikroskop altında sclera'nın, episcleral katman, stroma ve de lamina fusca sclera'dan oluştuğu görüldü. Mikroskobik incelemeler yapıldığında, anterior uvea'nın iris ve corpus ciliare'den, posterior uvea'nın ise choroidea'dan oluştuğu görüldü. Makroskobik olarak choroidea'nın posterior kısmında yer alan tapetum lucidum'un mavi-sarı renkte, üçgen şeklinde olduğu, alt kenarının discus n.optici'ye hafifçe temas ettiği ancak discus'u içine almadığı gözlemlendi. Işık mikroskobik olarak bu bölgenin içerdiği kollajen liflerden ötürü fibröz karakterde olduğu ortaya kondu. Corpus ciliare'de, her bir proc.ciliaris'in, stroma ve kan damarlarını çevreleyen 2 katlı epitelden oluştuğu görüldü. Bunlardan dışta yer alan katman pigmentsiz kübik epitelden, içte yer alan katman ise yoğun koyu pigmentli kübik epitel hücrelerinden oluşmaktaydı. Taramalı elektron mikroskop altında üç yada dört plicae ciliaris'lerin birleşerek proc.ciliaris'leri oluşturduğu görüldü. Plicae ciliaris'leri kendi aralarında birbirlerine bağlayan daha kısa ve zayıf ipliklerin olduğu da tespit edildi. Hem makroskobik hem de ışık mikroskobik olarak iris'in pupillar sınırının dorsal kesiminde corpora nigra'nın varlığı gözlemlendi. Mikroskobik olarak incelendiğinde, camera anterior bulbi'nin periferinde bulunan angulus iridocornealis'in sinus ciliaris, plexus aquosus angularis, lig.pectinatum ve trabeküler ağdan oluştuğu görüldü. Sonuç olarak bulbus oculi'nin yapısal özelliklerinin tanımlanmasının gelecekte göz üzerinde yapılacak olan morfolojik, fizyolojik ve patolojik çalışmalara ve de operatif girişimlere yardımcı olacağı düşünülmektedir.

In this study, it was aimed to reveal the morphological and morphometric properties of the eye of the cattle using microscopic methods. In order to perform this study, a total of 40 eyes of 20 cattle left and right were extracted without damaging surrounding tissues. Morphological images of the eyes were photographed under stereomicroscope. External morphometric measurements were obtained before the fixation step. Separate sections were taken from each eye detected after dissection to examine under both electron and light microscope. In order to perform light microscopic examinations, tissue samples were embedded in paraffin and 5-micrometer thick sections were applied with Crossmann's triple staining technique.Anterior 1/6 of the tunica fibrosa bulbi was formed by the cornea and the rest by the sclera. The epithelium anterius cornea covering the anterior face of the cornea consisted of a multilayer squamous non-keratinized epithelium. Under the electron microscope, cornea showed a wavy appearance due to the dense collagen fibers contained in the substantia propria layer. Under the light microscope, the sclera was composed of the episcleral layer, the stroma and the lamina fusca sclera.Microscopic examinations revealed that the anterior uvea consisted of iris and ciliary body and the posterior uvea consisted of choroid. Macroscopically, it was observed that the tapetum lucidum in the posterior part of the choroid was blue-yellow-green, triangular in shape, the lower edge of the optic disc was slightly in contact with this disc but did not contain it. Light microscopically, this region was found to be fibrous due to the collagen fibers it contained. In the corpus ciliare, each ciliary process was found to consist of a two folded epithelium surrounding the stroma and blood vessels. The outer layer consisted of non-pigmented cubic epithelium and the inner layer consisted of dense dark pigmented cubic epithelial cells. Under the scanning electron microscope, it was observed that three or four ciliary plicae merged to form ciliary process. It was also found that there were shorter and weaker bands that interconnected the ciliary plicae. Both macroscopically and light microscopically, the presence of corpora nigra was observed in the dorsal part of the pupillary border of the iris. Microscopically, the iridocorneal angle in the periphery of the anterior chamber was composed of ciliary cleft, angular aqueous plexus, pectinate ligament and trabecular meshwork.In conclusion, it is thought that identification of structural features of bovine eye will help future morphological, physiological and pathological studies and operative interventions.
