Discovery and characterization of nanobodies against the transcriptional repressor LexA of Pseudomonas aeruginosa

Abstract

Discovery and characterization of nanobodies against LexA of Pseudomonas aeruginosa. LexA is a transcriptional repressor that undergoes an autocatalytic cleavage induced by the protein RecA. This mechanism allows the derepression of those genes that are expressed in response to DNA damage. Their activity is highly mutagenic and for this reason can cause antibiotic resistance appearance. The main goal of this thesis is to characterize nanobodies directed against LexA to inhibit its autocatalytic cleavage, involved in genetic variability occurence, in order to reduce the induction of sos response.
