Abstract

The analyses of RAPD (Randomly Amplified Polymorphic DNA) and SRAP (Sequence Relatad Amplified Polimorphism) marker were carried out on 13 cultivars representing the two ecological races of avocado (Persea americana Mill.) and 7 cultivars of Mexican and Guatemalan hybrids. Eigtheen RAPD primers produced 116 DNA fragments in RAPD assay at the avocado cultivars. Twenty one SRAP primer combination produced 122 bands. A jacard's similarity coefficient matrix was generated and a dendogram was constructed using UPGMA (Unweighted Pair-Group Metod of Aritmetic Averages) cluster analysis for RAPD and SRAP analysis. Average similarity between genotypes ranged from %46 to %86 with RAPD marker and ranged from %49 to %90 with SRAP marker. The results obtained indicate that the SRAP and RAPD markes used are highly informative and are discussed in term of their implications for avocado germplasm characterization. Tree and leaf characteristics such as tree vigor, tree length, canopy of tree, tree shape, leaf length, color of shoots, leaf shape etc. are evaluated by using IPGRI standarts for twenty avocado cultivars. All quantitative morphological characters analysed by using SAS program.

Avokadonun iki ırkını temsil eden 13 çeşit ile Meksika Guatemala melezi olan 7 çeşit SRAP ve RAPD markırları ile analiz edilmiştir. On sekiz RAPD primerinden 116 bant; 21 SRAP primerinden 122 bant elde edilmiştir. RAPD ve SRAP analizlerinde Jacard'ın benzerlik matriksi kullanılmış ve UPGMA analizi ile dendogram oluşturulmuştur. RAPD'ın genotipler arasındaki benzerlik derecesi %46 ile %86 iken SRAP da bu değerler %49 ile %90 olarak hesaplanmıştır. Elde ettiğimiz sonuçlar avokado genotiplerinin karakterizasyonunda SRAP ve RAPD markırlarının kullanılabileceğini göstermiştir. Yirmi avokado çeşidine ait ağaç gücü, ağaç boyu, ağacın tacı, şekli, yaprak boyu, sürgün rengi, yaprak şekli gibi ağaç ve yaprak özellikleri IPGRI standartları baz alınarak incelenmiştir. Tüm morfolojik karakterler SAS bilgisayar programında analiz edilmiştir.
