Abstract

Bu çalışma kapsamında Apocrita alttakımını temsilen beş türün mitogenom verisi yeni nesil dizileme yaklaşımları ile üretilmiş ve biyoinformatik yaklaşımlar altında karşılaştırmalı olarak karakterize edilmiştir. Mitogenom uzunlukları Bombus lapidarius türünde 17,988 bç (total), Scolia hirta türünde 16,295 bç (kısmi), Collyria coxator türünde 15,933 (kısmi), Diplolepis fructuum türünde 15,102 bç (kısmi) ve Ormyrus tschami türünde 12,621 bç (kısmi) olarak belirlenmiştir. Bu türlerin mitogenom anatomilerinde tRNA ve protein kodlayan genlerinde (PKG) saptanan gen yeniden düzenlenme örüntüleri, Apocrita alttakımındaki sık gözlenme eğilimini desteklemiştir. A+T yönünde bir eğilim gözlenen bu alttakımda, zincir asimetri değerleri S. hirta türü dışında böcekler için önerilen genel model ile uyumlu bulunmuştur. PKG'lerin A+T içeriğinde beklenen örünütünün aksine C. coxator türünde nad4L, S. hirta türünde nad2 ve O. tschami türünde ise nad6 genlerinin %A+T içeriğinde önemli bir artış olduğu belirlenmiştir. nad4L (TTT) ve nad5 (GTG) genleri dışında, PKG'lerde başlama kodonu olarak ATN; sonlanma kodonu olarak TAA, TAG, TA− veya T− kodonlarının kullanıldığı tespit edilmiştir. İkincil yapısı inşa edilen rRNA genlerinin, rapor edilen Hymenoptera türlerininki ile uyumlu olduğu gözlenmiştir. Apocrita filogenisinin 14 süperfamilyayı temsilen 41 türün mitogenom verisi üzerinden yeniden inşa edilmesi sonucunda (i) Orussidae familyasının Apocrita alttakımının en yakın akrabalarını temsil ettiği; (ii) Ichneumonomorpha içerisinde yer alan Ichneumonidae ve Braconidae familyalarının kardeş grup ilişkisi; (iii) Trigonaloidea + Megalyroidea süperfamilyaları arasındaki kardeş grup ilişkisi; (iv) korunmuş bir Proctotrupomorpha ilişkisi; (v) Sphecoidea süperfamilyasının Apoidea'nin bazalında yer aldığı ve (vi) gerçek sosyal arıların bir monofili sergilediği belirlenmiştir.

In this study, mitogenomes data of five species representing the suborder Apocrita were generated by the new generation sequencing and are comparatively characterized under bioinformatics approaches. Mitogenome lengths have been determined as 17,988 bp (total) for Bombus lapidarius, 16,295 bp (partial) for Scolia hirta, 15,933 bp (partial) for Collyria coxator, 15,102 bp (partial) for Diplolepis fructuum and 12,621 (partial) bp for Ormyrus tschami. The gene rearrangement patterns detected in tRNA and protein coding genes (PKG) in mitogenome anatomies of these species supported the frequent tendency observed in suborder Apocrita. Chain asymmetry values were found to be consistent with the general model proposed for insects except for those of S. hirta in the suborder where a trend was observed in the A + T direction. In contrast to the expected pattern in the A + T content of PKGs, nad4L of C. coxator, nad2 of S. hirta and nad6 of O. tschami were found to be increase significantly. Except for nad4L (TTT) and nad5 (GTG) genes, ATN is used as a starting codon and TAA, TAG, TA- or T-codons are used as termination codons. The predicted secondary structures of rRNA genes were consistent with that of the reported Hymenoptera species. The recostructed Apocrita phylogeny based on 41 mitogenomes representing 14 super families concluded that: (i) the Orussidae family represents the closest relatives of the Apocrita suborder; (ii) supported sister group relationship of Ichneumonidae and Braconidae families as Ichneumonomorpha; (iii) sister group relationship between Trigonaloidea + Megalyroidea superfamily was recovered; (iv Proctotrupomorpha was recovered as monophyletic; (v) Sphecoidea and Apoidea were always found as sistergroup and (vi) eusocial bees were recovered as monophyletic.
