Abstract

Supervisor of the master's thesis: Dr. Maksim Bratčikov Number of pages: 42 Number of tables: 3 Number of pictures: 13 Originally considered low pathogenic, Acinetobacter baumannii in recent years has become one of the most difficult to treat pathogens causing nosocomial infections worldwide. Rapid adaptation of the genome in the processes of removal and degradation of antimicrobal treatment complicates the treatment of infections, so one of the most important goals in modern medicine to combat diseases caused by A. baumannii is the study of the bacterial genome. In this work, 376 A. baumannii isolates collected in 2018-2019 were investigated. in the microbiology laboratory. Methods applied to the research: selection of isolates based on EUCAST epidemiological thresholds, culture freshening, culture freezing, culture cultivation, lysate preparation, selection of gene primers for real-time PCR. Appropriate samples were prepared during the study to identify resistance genes in A. baumannii isolates, and primers suitable for identification and evaluation of carbapenemase VIM genes and colistin MCR-1, MCR-6, MCR-7, MCR-8, MCR-9 genes were determined.

Magistro darbo vadovas: Dr. Maksim Bratčikov Puslapių skaičius:42 Lentelių skaičius: 3 Paveikslėlių skaičius: 13 Acinetobacter baumannii bakterijos, iš pradžių laikytos mažai patogeniškomis, pastaraisiais metais tapo vienu iš sunkiausiai gydomų patogenų, sukeliančiu hospitalines infekcijas visame pasaulyje. Spartus genomo prisitaikymas antimikrobinių medžiagų šalinimo bei skaidymo procesuose komplikuoja infekcijų gydymą, todėl vienas iš svarbiausių tikslų šiuolaikinėje medicinoje siekiant kovoti su A. baumannii sukeliamomis ligomis – bakterijų genomo tyrimas. Šiame darbe buvo tiriami 376 A. baumannii izoliatai, surinkti 2018-2019m. mikrobiologijos laboratorijoje. Tyrimams pritaikyti metodai: izoliatų atrankos remiantis EUCAST epidemiologinėmis slenkstinėmis vertėmis, kultūrų šviežinimas, kultūrų šaldymas, kultūrų auginimas, lizatų paruošimas, genų pradmenų parinkimas realaus laiko PGR analizei. Tyrimo metu buvo paruošti tinkami mėginiai, kurie padėjo identifikuoti atsparumo genus A. baumannii izoliatuose, nustatyti karbapenemazių VIM genų ir kolistino MCR-1, MCR-6, MCR-7, MCR-8, MCR-9 genų identifikavimui ir įvertinimui tinkami pradmenys.
