Abstract

La jerdostatina es una desintegrina corta con motivo RTS que fue descubierta a partir de una librería de ADNc de la glándula de veneno de Protobothrops jerdonii. Su motivo de unión RTS (Arg, Thr, Ser) le permite reconocer específicamente a la integrina α1β1, bloqueando su interacción con el colágeno IV de la membrana basal. Como receptor del colágeno, la integrina α1β1 media la adhesión y migración celular, por lo que la jerdostatina se ha propuesto como un potencial agente antiangiogénico y antitumoral. Sin embargo, hasta ahora no se habían realizado ensayos en células del sistema inmune. Las células dendríticas, son las células presentadoras de antígeno por excelencia. Además de actuar como células centinelas, son fundamentales para la activación de linfocitos T vírgenes, activando y dirigiendo la respuesta inmune adaptativa, pero también son las encargadas de mediar la tolerancia inmunológica. a presente tesis de Maestría tuvo como objetivo la producción recombinante de la jerdostatina y evaluar su posible efecto inmunomodulador sobre la maduración y activación de las células dendríticas, con el fin de avanzar en la investigación de su posible aplicación terapéutica. Durante el desarrollo de esta Maestría se logró expresar la jerdostatina en E. coli, como proteína de fusión, que luego fue escindida por proteólisis. Se obtuvo un rendimiento de 0,56 mg/L de cultivo, un valor esperado según lo reportado en la literatura. Mediante inmunoensayos se comprobó que la jerdostatina recombinante fue capaz de unirse a su ligando, la integrina α1β1. Sin embargo no pudimos confirmar su capacidad de bloquear la unión entre la integrina α1β1 y el colágeno IV. Al evaluar su potencial inmunomodulador, la jerdostatina recombinante demostró la capacidad de inhibir la expresión de las moléculas coestimuladoras CD40 y CD86, además de MHCII y citoquinas proinflamatorias inducidas por ligandos de TLR, en células dendríticas. Por lo tanto, nuestros resultados indican que la jerdostatina inhibe la maduración de las células dendríticas en las condiciones estudiadas. Este trabajo revela entonces, un nuevo efecto de la jerdostatina, en vistas de su potencial aplicación como agente terapéutico.
