Mejora del índice de multiplicación in vitro de dos clones de boniato

Abstract

El aislamiento de ápices meristemáticos mediante cultivo in vitro es una técnica clave para eliminar patógenos especialmente relevante y utilizada en especies de reproducción vegetativa, como el boniato para obtener clones saneados. Sin embargo, es un proceso delicado que requiere formación y experiencia previa por el personal que lo realiza. Así, una vez se ha obtenido la planta saneada, es necesario rentabilizar al máximo su multiplicación. Con dicho fin, se requiere investigar los diferentes medios de cultivo que nos aseguren la supervivencia de las plantas saneadas y obtener el máximo índice de multiplicación en las fases posteriores, como la proliferación. El objetivo del presente trabajo fue estudiar el efecto del genotipo de boniato y de las citoquininas en la multiplicación in vitro de segmentos nodales previamente saneados mediante el aislamiento de meristemos. El material vegetal seleccionado fueron dos clones de boniato de fenotipos muy diferentes entre sí; uno comercial de piel naranja denominado “Rojo California”, y uno tradicional de la Comunidad Valenciana de carne blanca, conocido como “Blanco”. En cuanto a las citoquininas, se testó el efecto de la zeatina (ZT) y la kinetina (KIN) ambas de origen natural, y como sintéticas se utilizó la 6- bencilaminopurina (BAP), el 2-isopenteniladenina (2iP) y el thidiazuron (TDZ). Para ambos clones de boniato, los mejores índices de multiplicación se consiguieron con KIN o con el medio sin reguladores, con los que se consigue un índice de multiplicación de 3 en un ciclo de 6 semanas. Estos resultados resaltan que se puede rentabilizar la producción de plantas de boniato saneadas in vitro mediante la elección de citoquininas de origen natural, como la KIN, o incluso sin la utilización de reguladores, recortando así los gastos de producción y de obtención de dichas plantas.
