Abstract

Büyük ekonomik kayıplara neden olan mastitis, çeşitli sebeplerden meydana gelen meme dokusunun yangısıdır. Mastitisin bakteriyel etkenlerinden olan Klebsiella türleri aynı zamanda doğada yaygın olarak görülen fırsatçı bir patojendir. Enfekte olan canlıya antibiyotik tedavisi uygulanması gerekmektedir. Bu yüzden hastalık karşısında Klebsiella türlerinin antibiyogramı gerçekleştirilmeli ve antibiyotik dirençliliğine göre tedavisi yapılmalıdır.Çalışmamızda Aydın merkez ve periferdeki 162 inekten alınan süt numunelerinden bakteri izolasyonu ve identifikasyonu gerçekleştirildi. 67 adet gram negatif bakteri teşhis edildi. 67 suştan Klebsiella şüpheli olanların selektif besiyerlerinde izolasyonu yapıldı. İdentifiye edilen 23 Klebsiella spp.'nin 2'si Klebsiella ozaenae, 9'u Klebsiella oxytoca, 12'si ise Klebsiella pneumoniae olduğu tespit edildi. İdentifiye edilen Klebsiella türlerinin 19 farklı antibiyotiğe karşı dirençliliği test edildi. 23 adet izolatın genotiplendirmesi amacıyla ERIC-2 primeri kullanılarak gerçekleştirilen RAPD-PCR sonucunda 11 farklı genotip tespit edildi. Filogenetik analiz sonucunda genotiplerin %34-73 arasında benzerlik gösterdiği belirlendi.Anahtar Kelimeler: Genotiplendirme, İzolasyon, Klebsiella, Mastitis.

Mastitis, which causes great economic losses, is the inflammation of udder tissue consisting of various reasons. Klebsiella spp., one of the bacterial agents of mastitis, is also an opportunistic pathogen in nature. Antibiotic therapy should be applied to infected animals. Therefore, an antibiogram of Klebsiella spp. should be performed against the disease and treated according to antibiotic resistance.In this study, bacterial isolation and identification were made from milk samples taken from 162 cows in the center and periphery of Aydın. 67 gram negative bacteria are diagnosed. 67 strains of Klebsiella suspicion are isolated in selective media. 2 Klebsiella ozaenae, 9 Klebsiella oxytoca, and 12 Klebsiella pneumoniae are identified in the 23 Klebsiella spp. The resistance of the Klebsiella spp. to 19 different antibiotics is tested. For the genotyping of 23 isolates, 11 different genotypes are detected by RAPD-PCR using ERIC-2 primer. It was determined that the genotypes were similar between 34-73% in the phylogenetic analysis results.Key words: Genotyping, Isolation, Klebsiella, Mastitis.
