Identificación de fitoplasmas en cerezo (Prunus avium L.).

Abstract

Con la técnica de reacción en cadena de la polimerasa (PCR) se detectaron fitoplasmas en el floema de estacas de cerezo (destinado a la industria de transformación) procedentes de plantas con bajo crecimiento y decaimiento generalizado. Para el PCR directo se han utilizado los partidores P1/P7 y sobre el producto de amplificación de estos partidores se han realizados los PCR anidados con F1/B6 y R16F2/R2, que reconocen secuencias específicas del operón de genes ribosomales de los fitoplasmas. Con las enzimas de restricción TruI y HhaI se obtuvo un perfil característico del grupo ribosomal 16SrIII (grupo del X-disease). También se ha secuenciado el producto de amplificación de R16F2/R2 y con el análisis de la secuencia nucleotídica se confirmó que el fitoplasma encontrado en los cerezos analizados pertenece al subgrupo 16SrIII-J, en cuanto presenta el 99% de similitud con “Chayote witches’ broom” detectado en Brasil, “Chinaberry yellows” en Bolivia y “Delphinium phytoplasma” en el Reino Unido
