Abstract

It is well known that, thymol and heterocycles existing in nature are the most important sources for the prevention and treatment of various diseases and these are the starting point of many researches. The usage of pure compounds have become more popular in treatment of hardly curable diseases such as cancer as a main treatment or as apart of many treatment. Thymol (2-isopropyl-5-methylphenol) is a monotherpene isolated from plants of the Lamiaceae family (Thymus, Ocimum, Origanum and Monarda). Thymol and its derivatives have been studied widely due to their significant biological effects. It has biological activities such as, anticancer, antifungal, antibacterial and antioxidant activities. Benzothiophene and indole derivatives are an important group of heterocyclic compounds that have received great deal of attention in recent years due to their pharmacological activities. The cytotoxic and anti-carcinogenic effects of newly synthesized five different benzothiophene-thymol and indole-thymol hybrid molecules on human lung carcinoma (A549), human prostate carcinoma (LNCaP), human colorectal adenocarcinoma (Caco-2), and human breast adenocarcinoma (MCF-7) on various cancer cells were investigated in this study. In this context, the effective doses (EC50) in different cancer cells were calculated for two derivatives (AŞ1 and AŞ7) and their cytotoxic effects were evaluated. The effects of these two active derivatives on apoptosis-related genes’ (BAX, BCL-2, CASP3, CASP8, CASP9) mRNA levels were determined by qRT-PCR. While, the mRNA levels of BAX, CASP3, CASP8, CASP9 genes increased significantly compared to control, a decrease in BCL-2 mRNA level was detected. Image cytometry results showed that apoptosis was induced as a result of the administration of these two derivatives. In the light of all these results, it has been revealed that the new benzothiophene-thymol (AŞ1) and indole-thymol derivative (AŞ7) cause apoptosis induction by using two different mechanisms and exert cytotoxic effects. All these results showed that these two derivatives may have the potential to be used in cancer treatment.

Bilindiği üzere, doğada var olan timol ve heterosiklikler, çeşitli hastalıkların önlenmesi ve tedavi edilmesi için en önemli kaynaklardır ve araştırmaların da çıkış noktalarını oluşturmaktadırlar. Özellikle kanser gibi tedavisi zor olan hastalıklarda gerek ana tedavi gerekse ana tedavinin yardımcısı olarak, çeşitli etken maddelerin kullanımı popülerleşmektedir. Timol (2-izopropil-5-metilfenol), Lamiaceae familyasına (Thymus ,Ocimum ,Origanum ve Monarda ) ait bitkilerden izole edilen monoterpen fenoldür. Timol ve türevleri önemli biyolojik etkilerinden dolayı geniş bir literatüre sahiptir. Timol, antikanser, antifungal, antibakteriyel, antioksidan gibi biyolojik aktivitelere sahiptir. Benzotiyofen ve indol türevleri ise son yıllarda sahip oldukları farmakolojik etkilerden dolayı üzerinde yapılan çalışmaların oldukça arttığı önemli bir heterosiklik bileşik grublarıdır. Bu çalışma kapsamında yeni sentezlenen beş adet benzotiyofen-timol ve indol-timol hibrid moleküllerinin sitotoksik ve anti-karsinojenik etkilerinin insan akciğer karsinoma (A549), insan prostat karsinoma (LNCaP), insan kolorektal adenokarsinom (Caco-2) ve insan meme adenokarsinoma (MCF-7) gibi farklı kanser hücreleri üzerinde araştırılması amaçlanmıştır. Bu bağlamda farklı kanser hücrelerinde etkin dozları (EC50) iki türev için (AŞ1 ve AŞ7) hesaplanmış ve sitotoksik etkileri ortaya konulmuştur. Bu iki aktif türevin apoptoz ile ilişkili genlerin (BAX, BCL-2, CASP3, CASP8, CASP9) mRNA seviyelerine olan etkileri qRT-PZR ile belirlenmiştir. BAX, CASP3, CASP8, CASP9 genlerinin mRNA seviyesinde değişimler kontrole göre kıyasla önemli ölçüde artış gösterirken, BCL-2 mRNA seviyesinde ise azalış saptanmıştır. İmage sitometre sonuçları apoptozun bu iki madde uygulanması sonucunda indüklendiğini göstermiştir. Tüm bunların ışığında, yeni benzotiyofen-timol (AŞ1) ve indol-timol türevinin (AŞ7) iki farklı mekanizma kullanarak apoptoz indüksiyonuna neden olduğu ve sitotoksik etki gösterdiği ortaya çıkarılmıştır. Tüm bu sonuçlar, bu iki türevin kanser tedavisinde kullanılma potansiyeli olabileceğini göstermiştir.
