Abstract

Small cell lung cancer (SCLC), which accounts for 15-20% of all lung cancer types, is an aggressive malignancy with an extremely high mortality rate. The most frequently altered genes in SCLC are the tumor suppressor genes TP53 and RB1 and the MYC family oncogenes MYC, MYCL and MYCN. Overall, MYC genes are amplified and overexpressed in 20% of SCLCs, showing that MYC proteins are strong candidates as therapeutic targets in SCLC. MYC acts as a transcription factor that coordinates many biological processes.In this study, at first two lentiviral vectors for the constitutive (pCDH-MYC) and the inducible (pTIJ-MYC) expression of MYC and a lentiviral vector for the inducible down-regulation of MYC (pTRIPZ-MYC) were constructed. Subsequently, we infected MYC-non amplified SCLC cell lines, H209, with pCDH-MYC and H345 with pTIJ-MYC, and two MYC-amplified and over-expressing SCLC cell lines, N417 and H82, with pTRIPZ-MYC. Stable infected cells were selected with puromycin and for the inducible expression systems, MYC was over-expressed or down-regulated by addition of doxycycline. The effects of MYC over-expression or down-regulation on cellular proliferation, metabolism and gene expression were evaluated by western blot, qRT-PCR, miRNA/mRNA microarray (Affymetrix miRNA 4.0/ Illumina HT-12) and LC-MS analysis.Cellular proliferation was found to be dependent on the expression of MYC on the cells used in this study. Then, we evaluated the expression of miRNAs (miR-9, miR-181, miR-378, etc) and mRNAs (HK2,GLS1,LDHA,etc) that play an important role in cancer progression. We observed alterations in the expression of several MYC-regulated metabolic genes. The functions of these genes were analyzed by performing LC-MS analysis.Our results indicate that SCLC cells with MYC amplification are addicted to MYC for their survival. In our belief, this study highlights the relavance of Myc-regulated metabolic pathways for the development of future therapeutic strategies in SCLC.

Akciğer kanserlerinin %15-20'ni oluşturan küçük hücreli akciğer kanseri (KHAK) yüksek ölüm oranı ve agresif fenotiple karakterizedir. KHAK'lerinde en sık gözlenen genetik değişimler tümör baskılayıcı TP53 ve RB1 ile MYC ailesi onkogenlerde gözlenen değişimlerdir. Myc geni KHAK olgularının yaklaşık %20'sinde aşırı ifade ya da amplifikasyonu gözlenmektedir. Bu sebeple Myc geni KHAK tedavisi için önemli bir hedeftir. Myc pek çok biyolojik süreci düzenleyen önemli bir transkripsiyon faktörüdür.Çalışmamızda ilk olarak kalıcı (pCDH-MYC) ve indüklenebilir (pTIJ-MYC) ekspresyon vektörleri ile indüklenebilir baskılama vektörleri (pTRIPZ-MYC) tasarlandı. Ardından, Myc ampliikasyonu taşımayan H209 hücreleri pCDH-MYC ile H345 hücreleri pTIJ-MYC ile, Myc amplifikasyonu taşıyan H82 ve N417 hücreleri ise pTRIPZ-MYC ile infekte edildi. Kalıcı infekte edilmiş hücreler puromisin ile seçildikten sonra indüklenebilir vektör sistemleri doksisiklin ile muamele yapılarak Myc ifadesi düzenlendi. Myc ifadesindeki değişimin hücre çoğalması, metabolizma ve gen ekspresyonu üzerine etkisi western blot, gerçek zamanlı PZR, miRNA/mRNA mikrodizin (Affymetrix miRNA 4.0/ Illumina HT-12) ve LC-MS analizi ile araştırıldı.Çalışmada kullanılan hücrelerdeki hücresel proliferasyonun MYC ekspresyonuna bağımlı olduğu belirlendi. Sonrasında, kanser progresyonunda önemli rol oynayan miRNA'ların (miR-9, miR-181, miR-378, vb) ve mRNA'ların (HK2,GLS1,LDHA,vb) ekspresyonları araştırıldı. MYC tarafından düzenlenen metabolik genlerin ekspresyonlarında değişimler saptandı. Bu genlerin fonksiyonları LC-MS analizi ile değerlendirildi.Sonuçlarımız, MYC amplifikasyonuna sahip KHAK hücrelerinin sağkalımı için MYC'e bağımlı olduklarını göstermiştir. Bu çalışmanın KHAK'lerinde ileriki terapötik stratejilerin gelişiminde MYC tarafından düzenlenen metabolik genlerin önemini vurguladığı kanısındayız.
